As he reveals to Taiju the origin of the cave’s nitric acid, Senku sums up in one line what is actually a complex biochemical chain: “It’s miracle water, thanks to bat guano.” An apparent author’s shortcut, but actually the most efficient possible explanation of the process that supplied humanity with its nitrate for 2,000 years.

The biochemical chain

Guano is a complex mix of droppings, urine, runoff water, and prey remains that builds up under bat or seabird colonies. Its composition is rich in nitrogen, mainly in the form of urea (CO(NH₂)₂) and uric acid (C₅H₄N₄O₃) [Wikipedia] .

On contact with moisture, these molecules hydrolyze rapidly into ammonia (NH₃), which dissolves in the cave water as ammonium (NH₄⁺). That’s where the bacteria take over.

Two bacterial genera work in relay. First, Nitrosomonas (a chemolithoautotroph, it draws its energy from oxidizing ammonium) catalyzes:

NH₄⁺ + 1.5 O₂ → NO₂⁻ + 2 H⁺ + H₂O

Then Nitrobacter (or Nitrospira in some environments) takes over on the nitrite:

NO₂⁻ + 0.5 O₂ → NO₃⁻

This full chain, the oxidation of ammonium into nitrate by bacterial action, is called nitrification [Wikipedia] . It occurs in every aerated soil on Earth and is a key step of the nitrogen cycle .

Why acidic caves

In an environment acidified by dissolved atmospheric CO₂, the nitrate produced pairs with a proton to form dilute HNO₃ (typically 0.1 to 5% concentration, enough for a primitive alchemist to distill and concentrate). The HNO₃ slowly drips from the ceiling and pools on the floor. Over centuries, natural reservoirs of nitric acid form in certain caves with dense bat populations (Mexico, Texas, Southeast Asia).

The same mechanism produces saltpeter (potassium nitrate, KNO₃) elsewhere, when the potassium ion dominates in the environment (plant ash, manure). Saltpeter is the historical raw material of black powder [Wikipedia] .
